Hemagglutination Inhibition Asssay
The serum samples of the intranasally immunized mice after third immunization (bleed 2) were chosen for HIA. The HIA titer is the reciprocal of the highest dilution of serum exhibiting a complete HI. The median of titers of each group have been calculated and it was observed that the mice from group 2 and group 4 have significant HIA titers (≥40) which is considered to be associated with protection against influenza. Samples from group 2 and 4 of the mice that were intranasally immunized have also shown cross protection against the A/H1N1/Brisbane strain with HIA titers (320). The serum samples of the mice from second experiment (intranasal and sublingual vaccination) couldn’t show any HIA titers (<10) inferring a poor immunogenicity.
