Effect of induced T regulatory cells (Tregs) in mercury induced Autoimmunity (HgIA)

Autoimmunity is the immune response against self-antigens in healthy individuals in which self-reactive T cells were activated, which induces tissue destruction.  It was the third major cause of mortality. Autoimmunity can be caused by heavy metals like mercury, silver and gold. Type 1 diabetes, thyroid diseases, Systemic lupus erythematosus (SLE), scleroderma and Sjogren’s syndrome were some of the main types of autoimmune diseases. Heavy metal induced auto immunity through the production of anti nucleolar antibodies. It has been studied intensely using mice as the model organism. Sub toxic level of heavy metals like mercury or silver on susceptible mice can produce autoimmune responses.  T-regulatory cells can suppress the autoimmune responses. Here, we were investigating the effect of Tregs on mercury induced Autoimmunity.  

Interferon gamma knocked out B10.S mice were used as donor and recipient were the wild type. Interferon gamma mice were used to study the effect of interferon gamma on production of ANoA. Indirect immunofluorescence was performed for detection of serum antinucleolar antibodies (IgG1, IgG2a, IgG2b and IgG3) and polyclonal B-cell activation was measured by anti-ssDNA and anti-DNP antibodies using ELISA. Furthermore, level of IgG1 concentration was obtained by ELISA method.
